Peanut allergy is a serious chronic condition which has no cure.

The HILO study (High-Low Oral Immunotherapy) aims to compare the effectiveness of high dose probiotic peanut oral immunotherapy (PPOIT) and high dose peanut oral immunotherapy (no probiotic) versus low dose peanut oral immunotherapy for the remission of peanut allergy.

The HILO study (High-Low Oral Immunotherapy) aims to compare the effectiveness of high dose probiotic peanut oral immunotherapy (PPOIT) and high dose peanut oral immunotherapy (no probiotic) versus low dose peanut oral immunotherapy for the remission of peanut allergy.

Overview

Australia has the highest reported rates in the world, with 3 per cent of children affected by the condition. As there is currently no cure, peanut allergy management is focused on avoiding the allergen. There is an urgent need to find an effective peanut allergy treatment that improves quality of life and prevents reactions, hospitalisations and deaths.

Study Information

All children in the study will receive active peanut oral immunotherapy to be taken daily over 18 months. Your child will be randomised to one of three treatment groups.

  • High-dose peanut OIT combined with probiotic (HD PPOIT)
  • High-dose peanut OIT combined with probiotic placebo (HD OIT)
  • Low-dose peanut OIT combined with probiotic placebo (LD OIT)

Peanut food challenges will be completed before treatment and at the end of treatment to test for remission of allergy. Participants will also be followed up for a further 12 months after treatment.

Who can take part?

Children aged one to 10 years old who are living in Melbourne or Adelaide and have a peanut allergy confirmed by a food challenge.

What will you need to do?

  • Participants will be involved in the peanut OIT study for three years
  • Treatment involves daily peanut oral immunotherapy and probiotic or probiotic placebo for 18 months
  • Attend regular visits at your allocated hospital during the 18 months of treatment
  • Peanut food challenges that take place before and after treatment
